Veel van onze lichaamsprocessen vertonen 24-uurs fluctuaties die gedreven worden door een inwendige biologische klok. Deze fluctuaties zijn gezonde aanpassingen aan de dag-nacht wisselingen in de omgeving. Veroudering geeft verstoringen van 24-uurs ritmen wat kan leiden tot slaapproblemen. Recent is duidelijk geworden dat onderlinge communicatie tussen de zenuwcellen van de biologische klok van groot belang is om het gehele systeem goed te laten functioneren. In dit project onderzoeken we de manier waarop zenuwcellen communiceren en we testen we de hypothese dat deze communicatie verstoord raakt bij veroudering. De verkregen kennis zal gebruikt worden om verstoorde ritmen te verbeteren.

Nagenoeg al onze lichaamsfuncties vertonen 24-uurs ritmen. Deze worden geproduceerd door een inwendige biologische klok, die aan de basis van de hersenen ligt, in de suprachiasmatische kernen. Deze kernen bevatten hersencellen die genetisch geprogrammeerd zijn om ritmen met een periode van 24 uur te produceren. Het is van groot belang dat de ritmen die in de neuronen geproduceerd worden, gelijk lopen met elkaar. In de biologische klok van jonge, gezonde mensen is dit het geval. Ons onderzoek toont aan dat de communicatie tussen neuronen bij veroudering afneemt. Hierdoor zijn de neuronen op verschillende tijdstippen actief, en de populatie als geheel vertoont hierdoor een verzwakt ritme. Verschillende neurotransmitters zijn waarschijnlijk betrokken bij de onderlinge communicatie. Wij hebben een rol voor GABA en VIP kunnen vaststellen, en onderzoeken welke mogelijkheden er zijn om de communicatie te verbeteren. Hiervoor gebruiken wij ook muizen soorten die een versnelde veroudering laten zien. Een opmerkelijke bevinding is dat een verhoging van activiteit van de muizen, een verbetering van het neuronale netwerk van de suprachiasmatische kernen leidt.

Samenvatting van de aanvraag

Circadian rhythmicity is a fundamental feature of living organisms. In mammals, the suprachiasmatic nuclei (SCN) function as a major circadian pacemaker, or ‘clock’. Understanding the neuronal communication within the network of the SCN is of great importance for understanding the functioning of the circadian system, as well as for the treatment of a dysfunction of the clock in diseases. Aim: The present proposal focuses on the neuronal network properties of the clock, and on the mechanisms underlying cellular synchronization. We will also investigate the deterioration of the circadian system that occurs with aging. We hypothesize that an impairment of neuronal synchronization within the SCN contributes to the specific characteristics observed with aging, such as sleeping and alertness problems. Approach: We apply a broad range of methodological approaches, ranging from behavioral, extracellular in vivo and in vitro recordings, patch clamp and imaging recordings and real time clock gene (i.e. Per2) expression recordings. We also designed experiments to test neuronal networks in the SCN of naturally aged wild type mice and in conditional (SCN-specific) prematurely aging mice. Expected outcomes: In vivo experiments will show how desynchronization and waveform changes of the SCN affect the timing and amplitude of the sleep-wake rhythm, and the capacity of the clock to adjust to shifted cycles. In vitro experiments will reveal coupling mechanisms in the clock and the role of specific neurotransmitters and ionic channels. We will also gain insight on whether the identified coupling parameters are subject to the process of aging, and as such may contribute to age-related disease. Impact: Our hypothesis that decreased synchronization among SCN neurons contributes to aging of the circadian system is new and will shed light on the treatment of aging and potentially other circadian disorders. Importantly, the SCN neuronal network offers an excellent model for the investigation of age-associated desynchrony in the brain.
