The transproter of bacterial cell wall precursors: the ideal drug target.

Van oudsher is de opbouw van de bacteriële celwand hèt doel geweest voor antibiotica en de ontwikkeling hiervan. Door de toenemende antimicrobiële resistentie tegen penicillines en aanverwante antibiotica verliezen we langzaam onze grip op dit belangrijke doelwit.

Recent onderzoek heeft binnen de celwand een nieuw doelwit blootgelegd: de transporter van de celwandbouwstenen. Remmers van de transporter in de bacterie maken een goede kans om als nieuw antibioticum ingezet te worden.  

De onderzoekers ontwikkelen nieuwe methoden om remmers van deze transporter te identificeren. Zij testen een breed scala aan verschillende moleculen. Ook onderzoeken zij de meest geschikte moleculen op hun potentie om te worden ingezet als nieuw antibioticum.

Eindverslag

Dit project was gefocust op de ontwikkeling van nieuwe wijzen van zoeken naar nieuwe antibiotica die de, voor bacteriën essentiële, bacteriële celwand synthese remmen. De speciale aandacht kreeg het transport proces van de celwandsubeenheden over de plasmamembraan van de bacterie. Voor dit transport proces is het eiwit FtsW essentieel. Dit eiwit verzorgt het transport van de celwandsubeenheden die in de vorm van Lipide II via FtsW over de membraan worden getransporteerd. Er is en assay opgezet om de interactie (en de verstoring daarvan) van FtsW met Lipide II te kunnen meten. Deze assay zal binnenkort worden ingezet voor de screening van 30,000 compounds om zo nieuwe antibiotica (“lead compounds”) te identificeren die specifiek het transportproces verstoren. Voor de eventuele verdere ontwikkeling van de “leads” is het hebben van structuur informatie essentieel. Daarom werd parallel aan de ontwikkeling van de screeningsassay getracht de kristalstructuur van FtsW (of een homoloog daarvan) op te helderen. Hoewel een succesvolle methode voor de isolatie en zuivering van grote hoeveelheden eiwit werd ontwikkeld en ook kristallen konden worden gegroeid, bleken deze kristallen niet (voldoende) diffractie te geven waardoor de kristalstructuur niet kon worden opgehelderd. Verdere pogingen om tot goede kristallen te komen zullen in de komende tijd worden gedaan.

Samenvatting van de aanvraag

The bacterial cell wall synthesis machinery is a unique and accessible target for antibiotics. The central event in this biosynthesis pathway is the transport of cell wall subunits across the bacterial plasma membrane, which is catalyzed by an essential and highly conserved protein, FtsW, whose mechanism of action was recently identified by the Breukink group. The key role played by FtsW in cell wall biosynthesis and its presence in the genomes of all cell wall-bearing bacteria make it an ideal new target for antibiotic development. This international and multidisciplinary program aims at developing novel high throughput screening assays that will allow the identification of inhibitors of this essential transporter. We will make use of a library of 30,000 molecules available at the only academically owned compound library and high throughput screening facility of the McMasters University in Canada. In parallel, efforts aimed at crystallizing and solving the crystal structure of FtsW from a number of bacteria will be initiated, which will be essential for the further development of lead compounds identified in the inhibition screen. The strength of this project is reflected in its multidisciplinarity, involving groups that specialize in membrane biochemistry, synthesis/screening, and X-ray crystallography, as well as in the fact that clones of FtsW homologs from different bacteria will be tested and have already been cloned and purified. Together, these efforts will generate a plethora of important information on an essential process in the cell wall biosynthesis pathway, ultimately leading to the identification of novel antibiotics that will be further developed for deployment in the fight against resistant bacteria.
