How to maintain a stable genome? A Passenger perspective

Een opvallend kenmerk van kankercellen is hun afwijkend aantal chromosomen. Gezonde cellen hebben 46 chromosomen, terwijl kankercellen vaak meer of minder dan 46 chromosomen hebben. Dit komt omdat tijdens de celdeling vaak fouten worden gemaakt bij de verdeling van de zuster chromosomen over de twee dochtercellen. Een eiwitcomplex dat essentieel is voor het goed laten verlopen van die chromosoomscheiding is het zogenaamde Chromosomaal Passenger Complex (CPC). Het CPC verricht verschillende taken op diverse plekken in de delende cel. Hoe en waarom dit complex zo goed kan “multi-tasken” zijn de vragen die we hebben geprobeerd te beantwoorden in dit project. We hebben daartoe verschillende grootschalige biochemische experimenten uitgevoerd om nieuwe bindingspartners en substraten van het CPC te identificeren. Deze zouden kunnen verklaren waar, waarom, en hoe het CPC zijn functies in de cel uitvoert en zorg draagt voor chromosomale stabiliteit. Deze aanpak heeft een aantal interessante kandidaten opgeleverd, waarvan we vervolgens hebben onderzocht hoe zij de functie van het CPC in de cel reguleren.

Samenvatting van de aanvraag

Gaining a better understanding in a fundamental cellular process and identifying new targets for anti-cancer therapy; my proposal aims to combine both. Central to my research is the fact that, in marked contrast to normal cells, cancer cells have gained or lost entire chromosomes. Thus, analyzing cancer cells can lead the way to unraveling the regulation of chromosomal stability. In addition, mechanistic understanding of the factors that ensure chromosomal stability can result in new therapeutic strategies to selectively kill chromosomal unstable cancer cells. Work from my group already paved the way for this research line. We showed that the chromosomal passenger complex (CPC) controls several processes in the dividing cell that ensure faithful segregation of the duplicated chromosomes. Now I propose to reveal how this protein complex exerts its various functions in the dividing cell since it will provide essential knowledge on how chromosomal stability is ensured. In my research functional siRNA screens will be combined with biochemical and chemical-genetic approaches to identify new factors involved in the spatio-temporal regulation of the chromosomal passenger complex and to discover novel downstream targets. I expect to uncover novel functions of the CPC in signaling pathways governing genomic stability and to reveal new substrates of the complex involved in chromosome segregation. Moreover, their potential as anti-cancer target will be explored. Thus by unwiring the workings of the chromosomal passenger complex I expect to gain novel insights into the origin of chromosomal instability in cancer cells.
