Linking glycobiology to autoimmunity: towards a better understanding of the citrulline-specific immune response in rheumatoid arthritis

Het ziektebeeld reumatoïde artritis wordt gekenmerkt door ontsteking van de gewrichten waardoor schade en invaliditeit ontstaat. Het ziektebeeld ontstaat door een complex samenspel van verschillende factoren. Recent onderzoek heeft laten zien dat autoantistoffen tegen gecitrullineerde eiwitten (anti-citrullinated proteïne antistoffen, ACPA) een belangrijke rol spelen in het ziekteproces. Uit ons eigen onderzoek blijkt dat ACPA directe ziekte bevorderende eigenschappen hebben waardoor ontsteking mede veroorzaakt of versterkt kan worden. Het beoogde project heeft als doel aan ACPA gebonden suikerstructuren nader te onderzoeken en te karakteriseren. Het is onze hypothese dat kennis van deze suikerstructuren en hun biologische effecten de verklaring zal geven hoe ACPA bijdragen aan het ziekteproces.
