Een dynamisch en humaan mini long bioreactor als model om SARS-CoV2 infecties en therapie effecten te bestuderen

De COVID-19 pandemie heeft duidelijk gemaakt dat snel handelen belangrijk is om wereld gezondheidsproblemen adequaat te bestrijden. Naast het vroegtijdig opsporen en behandelen van besmettingen, is het belangrijk om systemen te hebben om infectie mechanismen te begrijpen en vaccins te ontwikkelen.

Kweken van menselijke longcellen

Dit project heeft als doel menselijke longcellen te kweken en daarbij de fysiologie van de longen zo veel mogelijk na te bootsen. Luchtweg ‘epitheel’ cellen en bloedvaten bekledende ‘endotheel’ cellen zullen tegelijk in een bioreactor worden gekweekt, en vormen een kunstmatige luchtweg en bloedvat, respectievelijk, waarlangs lucht en groeimedium stromen. Dit model systeem benadert de echte long beter dan de huidige beschikbare testmethoden, en kan virale infecties waarschijnlijk beter nabootsen. Met ons systeem hopen we veelbelovende medicijnen op een snelle manier te kunnen testen voordat deze bij patiënten kunnen worden ingezet. Bovendien kan de bioreactor op commerciële schaal gemaakt worden, waardoor andere laboratoria het makkelijk kunnen gebruiken.

Samenvatting van de aanvraag

Research question Can we develop an in vitro microfluidic human lung-on-a-chip model that mimics the in vivo lung physiology to (1) improve analysis of SARS-CoV2 host-virus interaction, (2) understand viral infection dynamics, and (3) develop anti-viral therapeutic strategies? Urgency The current pandemic of SARS-CoV2 shows the importance of quick responses: in the first place the health vigilant system to accommodate immediate health issues, but secondly the scientific community to develop viral tests and vaccines for improved monitoring and protection. The latter relies on dependable and physiological experimental models to quickly decipher the biology of the virus. Hypothesis A human lung on a chip functionally mimics the in vivo organ, is very suitable to delineate the biology of the virus and better predicts outcome of therapeutic agents. Moreover, knowledge of the virus and the inflicted pathology will provide better information to support guidelines and thus benefits society. Approach To improve modelling of interactions of SARS-CoV2 with mucociliary epithelium, viral infection dynamics, and development of therapeutic strategies, we aim to develop a reliable and reproducible human lung on a chip using exclusive and patented technology (300MICRONS). This miniaturized bioreactor consists of two individually perfuseable microfluidic compartments separated by a porous culture membrane. The membrane allows primary pulmonary epithelial cells (nasal, airway, alveolar) and endothelial cells to be cultured on either side of it. The compartments have independent fluid- and/or airflow, allowing epithelial cells to be air-exposed. Initially, epithelial monocultures are used, but the complexity will gradually increase with cocultures of epithelial- and endothelial cells to mimic the physiology of the airways. After establishing the cultures, SARS-CoV2 pseudoviruses will be applied to the bioreactor to monitor the virus-epithelium interactions, and the role of the endothelium.
