TissueFAXS: the equivalent to flow cytometry for multiparameter quantitative analyses in tissues

Projectomschrijving

Aan de ontwikkeling van ziekten (bijvoorbeeld atherosclerose, autoimmuunziekten en kanker) ligt vaak een verstoorde functie van cellen ten grondslag. Eiwitten spelen een belangrijke rol bij de functie van cellen en veranderingen in productie van eiwitten is dan ook vaak betrokken bij de ziekte ontwikkeling. Het is daarom van groot belang in staat te zijn te weten welke, en hoeveel eiwitten gemaakt worden door de verschillende cellen in het weefsel tijdens de ontwikkeling van een ziekte. In dit project wordt een geavanceerde microscoop gebruikt die ons in staat stelt de aanwezigheid en hoeveelheid van verschillende eiwitten in gezonde en zieke weefsels te meten. Op deze manier wordt er nieuwe informatie verkregen over welke eiwitten betrokken zijn bij het niet goed functioneren van cellen, wat belangrijk is voor de ontwikkeling van nieuwe therapieën om ziekten te voorkomen of te behandelen.

In dit project bestond de investering uit een geavanceerd microscopie systeem (de TissueFAXS®) dat gebruikers in staat stelde tegelijkertijd de expressie van verschillende eiwitten op een individuele cel kwalitatief en kwantitatief in kaart te brengen. Deze analyses konden worden uitgevoerd op hele dunne plakjes weefsel, gekweekte cellen, of bacterien met als doel meer inzicht te verkrijgen in de rol die verschillende eiwitten spelen tijdens normale functie van cellen of juist bij het ontstaan van ziekten. Zonder dit systeem waren dergelijke kwantitatieve analyses van eiwitexpressie in weefsels niet mogelijk om uit te voeren. De microscoop opstelling is gepositioneerd in het UMCG Imaging & Microscopy Center (UMIC) zodat het voor alle onderzoekers van UMCG, Rijksuniversiteit Groningen, maar ook daarbuiten [inter)nationaal] toegankelijk is.

Samenvatting van de aanvraag

With the advances in technologies of the last decade, a vast array of knowledge has been generated regarding cell behaviour in different conditions. More recently, it has become clear that microenvironments of cells in organs are essential contributors in creating pertinent niches that control cell (dys)function and pathogenesis of disease. Advances in array-based gene expression - and kinase activity profiling on whole tissue specimens - have significantly contributed to understanding of the cellular pathways involved in cell behaviour in both health and disease. Although data from these array-based studies definitely give clues about the genes and pathways that are active in various conditions, in-depth quantitative analyses at the protein expression level are required to validate array results and to gain insight into the level at which certain molecular pathways are controlled (e.g. at the gene- or at the protein expression level, posttranslational modifications). Generally, such quantitative protein expression studies are performed on whole tissue protein lysates ignoring the cellular context in which cells of interest reside. The TissueFAXS® system allows such quantitative multi-parameter analyses at the single cell level in tissue sections as well as in cultured cells, cell smears and cytospots. As quantitative analysis of protein expression at the single cell level in tissues is as yet not possible within our institution, but is considered an essential tool to strengthen studies employing complex tissues, we here apply for this equipment. The microscope-based TissueFAXS® system is the equivalent to flow cytometry in tissue. Users of the equipment applied for are PIs of research labs within the UMCG whose research is embedded in the research institute GUIDE, with focus on translational, innovative drug-oriented research in which molecular and cellular mechanisms underlying disease and responses to drug treatment are the common denominator. Analyzing protein expression levels quantitatively in tissues at the single cell level using the TissueFAXS® system will provide significant novel information about the control of cell dysfunction at the molecular level and the basis of success (and failure) of conventional as well as new classes of smart drugs, and will be of essential added value for local, national as well as international collaborative studies of the team of applicants.
