Surface plasmon resonance as tool in the analysis of biomolecular interactions that play a role in the etiology of cardiovascular disease.

Van de NWO investeringssubsidie, Middelgroot, is een uiterst gevoelig apparaat aangeschaft voor de meting van “oppervlakte plasmon resonantie” (SPR). Veranderingen in SPR ontstaan dan wanneer twee moleculen met elkaar een binding aangaan en het meten van SPR signalen stelt wetenschappelijk onderzoekers in staat om zeer nauwkeurig te bepalen of bepaalde moleculen met elkaar kunnen binden (en reageren) en hoe die binding plaatsvindt.De  investering heeft ertoe geleid dat bestaande interacties tussen (ziekmakende) menselijke moleculen beter zijn gekarakteriseerd, en daarnaast hebben de betrokken onderzoekers nieuwe interacties ontdekt die van belang kunnen zijn voor verschillende onderzoeksterreinen zoals dat van de hart- en vaatziekten, of dementie. De technologie wordt ook toegepast bij de ontwikkeling van nieuwe medicijnen. Hierbij bestuderen de onderzoekers de aard en sterkte van de binding tussen nieuwe kandidaat medicijnen en biomoleculen zoals die in het menselijk lichaam voorkomen. SPR meting helpt hierbij om nieuwe en verbeterde medicijnen te ontwikkelen.

Met behulp van de NWO investeringssubsidie, Middelgroot, is een uiterst gevoelig apparaat aangeschaft voor de meting van “oppervlakte plasmon resonantie” (SPR). Veranderingen in SPR ontstaan dan wanneer twee moleculen met elkaar een binding aangaan en het meten van SPR signalen stelt wetenschappelijk onderzoekers in staat om zeer nauwkeurig te bepalen of bepaalde moleculen met elkaar kunnen binden (en reageren) en hoe die binding plaatsvindt.
Gegeven het feit dat vrijwel elke reactie in ons menselijk lichaam het gevolg is van de binding van twee of meer moleculen aan elkaar, is het voor ons begrip van het ontstaan van ziekte van groot belang dat deze interacties bestudeerd kunnen worden. Binnen het Cardiovasculair Research Instituut Maastricht (CARIM) is dit SPR-apparaat nu geplaatst om, samen met andere technieken, ingezet te worden ter vergroting van het begrip van de hart-en vaatziekten en om te assisteren bij het ontwikkelen van nieuwe therapieen.
De investering heeft ertoe geleid dat bestaande interacties tussen (ziekmakende) menselijke moleculen beter zijn gekarakteriseerd, en daarnaast hebben de betrokken onderzoekers nieuwe interacties ontdekt die van belang kunnen zijn voor verschillende onderzoeksterreinen zoals dat van de hart- en vaatziekten, maar ook voor het gebied dat de dementie bestudeert. De technologie wordt verder nu ook toegepast bij de ontwikkeling van nieuwe medicijnen. Hierbij bestuderen de onderzoekers de aard en sterkte van de binding tussen nieuwe kandidaat medicijnen en biomoleculen zoals die in het menselijk lichaam voorkomen. SPR meting helpt hierbij om nieuwe en verbeterde medicijnen te ontwikkelen omdat ze een direct zicht biedt op het bindingsproces dat noodzakelijk voor de werking van de nieuwe (kandidaat) medicijnen.

Samenvatting van de aanvraag

A central phenomenon in biology is interaction between molecules resulting in a change of function of one or both of the binding partners. Ligand binding may thus generate new properties and/or initiate a change in a biological system which in itself may constitute a signal. Given the complexity of biological systems, interactions between proteins or between proteins and other ligands (e.g. metal ions, metabolites, DNA, lipid membranes, carbohydrates), are crucial to the majority if not to all biological processes and thus to the proper functioning of body fluids, cells and tissues. In today's life science research, notwithstanding the information that is encoded by the nucleotide sequence of the human genome, it is the study of proteins and protein interactions that is crucial to understand how proteins work in both health and disease. Several methods have been devised over the course of decades to study protein-ligand interactions. Commonly encountered problems comprise, however, the amount of proteins/analytes needed, the complexity and duration of experiments, the need for fluorescent or radioactive labeling of analytes, costs and lack of reproducibility. More recently, surface plasmon resonance (SPR) was introduced. In this technology a biospecific sensor chip is used that enables label-free detection of analytes binding to an immobilized target molecule and circumventing the before mentioned problems. SPR has successfully provided detailed and real-time quantitative information on the binding events that occur during complex biomolecular interactions and in this application we present pilot data that illustrate the unique suitability of SPR for our research objectives. The Cardiovascular Research Institute Maastricht (CARIM), recognized by the Royal Netherlands Academy of Arts and Sciences as a Research School on Cardiovascular Diseases, focuses on the humoral, cellular and vascular aspects of cardiovascular disease. In virtually all of these aspects protein-ligand binding is involved in homeostasis and pathology and the study of protein-ligand interactions contributes significantly to the molecular and mechanistic understanding of processes involved in the etiology of cardiovascular disease. The specific processes that we propose to study comprise interactions between (i) isolated proteins or domains thereof (ii) isolated proteins or protein domains on the one hand and small drug-like compounds on the other hand (after Virtual Ligand Screening) (iii) proteins (domains) and lipid (synthetic) membranes, (iv) ligand fishing (in combination with MALDI-TOF mass spectrometry) for the identification of binding partners (v) protein inhibition studies using synthetic peptides or antibodies (vi) antibody selection and characterization. We aim to exploit and combine surface plasmon resonance technology in an innovative proteomics research environment with state of the art technologies to enable discovery and quantitative description of protein-ligand interactions that underlie the molecular basis of cardiovascular disease. The technologies already available in our institute include kinetic analysis, protein engineering, recombinant DNA technology, protein purification techniques, structural bioinformatics analysis, virtual ligand screening based on high-throughput flexible docking, chemical protein synthesis and tandem MALDITOF mass spectroscopy
